Comparison of PCR and ELISA methods for detection of bovine leucosis in dried blood spots

Abstract

PCR and ELISA methods for detection of bovine leucosis in dried blood spots on porous membranes were compared. Dried samples were analyzed by real-time PCR using several diagnostic test-systems. 19 and 20 samples were detected as positive by PCR and ELISA, correspondently. 14 samples among them were positive in both methods. PCR and ELISA recovered 26 positive samples (47%) in total. The results of analysis of dried and native samples were in good agreement. Obtained results showed that whole blood sampling in a form of dried spots on membrane can be used as a convenient and reliable way for
biological fluid sampling for bovine leucosis screening.
Key words: enzootic bovine leucosis, ELISA, PCR, porous membranes, dried blood spots.
