dehydrogenase from Arabidopsis thaliana and soya: expression in E.
coli cells and kinetic properties of recombinant enzymes
It is the first case of
successful expression in E.coli cells of NAD+-dependent
formate dehydrogenases (EC 126.96.36.199) from plants Arabidopsis thaliana and
soya Gyicine max as active and soluble enzymes. Both enzymes were
obtained in homogeneous form and kinetic properties of recombinant enzymes were
studied. It was shown that new formate dehydrogenases have lower Km values
for coenzyme NAD+ and second substrate formate compared to ones for
the enzymes from bacteria and yeasts. Comparison of kinetic properties of plant
formate dehydrogenases with NAD+ and NADP+ has
shown that its coenzyme specificity is similar to one for the from bacterium
from Pseudomonas sp.101.
