The degradation of streptokinase by renal cathepsin B

Abstract

Cathepsin B was isolated from human kidneys and incubated at 37oC and pH 5.0 with streptokinase of the preparation Streptasa ("Belmedpreparat", Minsk). Cathepsin activity against streptokinase was studied with gel-filtration, PAAG-electrophoresis and with functional tests. The appearance of degradation products (8-10 kDa and about 50 kDa) was essential after 5 min of incubation but didn't manifest in any change of the preparation plasminogen-activating properties. Further streptokinase degradation caused first elongation of plasminogen activation and then inhibition of fibrinolysis up to its complete abrogation.
