The function of the human complement cascade after the proteolytic serum treatment in vitro.

Abstract

Proteolytic treatment of serum with pancreatic kallikrein leads to decrease of complement-mediated hemolysis velocity and reduction of hemolytic complement capacity (HCC), mainly in alternative pathway of complement activation (APC). It was demonstrated in experiments with purified proteins that changes showed caused by proteolytic degradation of C3 component. Decreasing of functional hemolytic activity was observed after treatment of C3 with elastase. C1-inhibitor prevents proteolytic splitting of C3 and increases the level of inhibitory threshold for kallikrein.
